High-performance Liquid Chromatographic Determination of Polyamines in Tomato Callus
本文用HPLC分离,内标法定量测定了蕃茄愈伤组织中的精胺、亚精胺、尸胺和腐胺。样品经DNS-Cl衍生和Sep-pak sillca短柱纯化。经μ-Bondapak NH2柱分离后,荧光检测。最低检出限达0.1～0.3p mol,添加回收率在89.1～102.1%,相对标准偏差小于2.3%。
Spermine,spermidine,cadaverine and putrescine in tomato callus were determined with 1,6-hexanediamine as internal standard.After DNS deriving,the sample was purified through Sop-pak silica cartridge,separated on μ-Bondapak NH2 column with mobile phase as chloroform:toluene:triothylamine(89:10:1,V/V)and detected by fluorescence detector at Ex=338 nm and Em=495 nm.The method showed good recovery in the range of 89.1～102.1%,the relative standard deviation of less than 2.3% and detection limit at 0.1 to 0.3 p mol.
